Yes. Two players can play together. Choose the 2-player world setting so guides open the two-player implementations where they exist.
4+ is a world configuration for groups of four or more. It is not a four-player maximum. Sessions still support up to 12 players.

Some challenges use different implementations for 2-player, 3-player, and 4+ worlds. Ring Room, for example, uses 4 slots in 2-player and 4+, and 3 slots in 3-player.
